How to improve time resolution in microdialysis?

A method has been developed for improved time resolution in on-line microdialysis. Conventional microdialysis sampling once every 10 or 20 minutes is not enough to describe a fast transmitter release accurately. In the new method analysis of dopamine (DA) and serotonin (5-HT) with 3 minute time resolution has been achieved.

The method is surprisingly simple: instead of using one 10 - 20 uL injection loop, a 14 port valve with 3 loops (between 1 - 4 uL) has been used. Each loop contains information of a unique time period.

Three parallel 50 x 1 mm HPLC columns are used for fast analysis of neurotransmitters. Run duration is shorter than filling time of the 3 loops. Handling data and conversion to response plots is done by automated procedures.

 

No autosampler is required in an on-line setup.
